Abstract
We developed an optical force clamp to apply loads to single molecules of kinesin moving on microtubule filaments. The effects of forward, backward, and sideways loads on kinesin motion suggest that the underlying reaction cycle contains a single working stroke that is well aligned with the direction of motion.
